Universidad Nacional de Córdoba, 2004 Summary: El éxito del tratamiento endodóntico depende de la eliminación de tejido pulpar, microorganisrnos, dentina afectada, y de la preparación del conducto radicular para permitir su obturación y sellado.El propósito de este trabajo fue evaluar, in vitro, el efecto del hipoclorito de sodio 2,5 porciento (NaOCL), EDTA 17 porciento, gluconato de clorhexidina 0,2 porciento (CHX) y agua destilada (testigo) sobre la Permeabilidad Dentinaria Radicular así como su Acción Antibacteriana.PERMEABILIDAD DENTINARIA RADICULARPenetración de colorante: Dientes unirradiculares fueron instrumentados, irrigados, llenados con azul de metileno 2 porciento, lavados y cortados en tercios. La permeabilidad dentinaria se midió empleando dos procedimientos. Medición lineal: se registró la máxima penetración del colorante en las cuatro caras de cada tercio. Medición de áreas: se midió la superficie coloreada en cada tercio, en imágenes digitalizadas. Con Friedman y Kruscall Wallis todas las soluciones mostraron mayor efecto permeabilizante en el tercio coronario, fuego medio y apical. Tuvo mayor efecto permeabilizante EDTA 17 porciento, luego NaOCL 2,5 porciento, CHX 0,2 porciento y agua destilada.Pasaje de iones. Dientes unirradiculares obturados con dos pastas de hidróxido de calcio. Se midió la concentración de OH- y Ca++ en el líquido circundante en tiempo inicial, 7, 14 y 30 días. ANOVA y Scheffé mostraron que con Ca(OH)2 más agua destilada, hubo incremento significativo don iones a los 14 y 30 días, siendo mayores los obtenidos con EDTA 17 porciento. Con Ca(OH)2 más propilenglicol no se observó aumento significativo de pH.ACCIÓN ANTIBACTERIANADifusión en agar con halos NaOCL 2,5 porciento = 21 mm. EDTA 17 porciento = 20 mm. CHX 0,2 porciento = 14mm. Dilución en agar.- NaOCL y CHX 0,2 porciento, EDTA menor de 5 porciento.Acción antibacteriana sobre la dentina. Se emplearon trozos radiculares apicales y rnedios. Fueron esterilizados, contaminados con Enterococcus faecalis, sumergidos en los irrigantes e incubados a 37 grados C. El recuento de células viables se realizó a 0, 4, 8 y 24 horas. ANOVA y Tukey mostraron mejor acción antibacteriana de NaOCL 2,5 porciento, luego CHX 0,2 porciento y EDTA 17 porciento.Summary: The success of endodontic treatment depends of plural tissue, microorganisms, affected dentine elimination as well as the right root canal shaping, filling and sealing.The aim of this work was to evaluate, in vitro, the effect of sodium hypochlorite 2,5 porcentage (NaOCL), chlorhexidine gluconate 0,2 porcentage (CHX), EDTA 17 porcentage and distilled water (control) on dentin permeability and its antimicrobial action.ROOT DENTIN PERMEABILITYDye penetration. human single root teeth were instrumented irrigated and filled with methylene blue dye 2 porcentage. Then the root canals were washed and cut into cervical, middle and apical thirds. The permeability of root dentin was measured using two methods. Lineal rneasurement the largest dye penetration in bucal, lingual, mesial and distal sides of the each third was evaluated. Area measurement in digitalized images, in each third the dye penetration was measured. The results using Friedman and Kruskall Wallis test showed all solutions highest permeability effect in coronal, then middle and apical third. The EDTA 17 porcentage induced a considerable increase in permeability, then NaOCL 2,5 porcentage, CHX 0,2 porcentage but no distilled water.Ion diffusion through root dentin. The teeth with single canals were instrumented and filled with two calcium hydroxide pastes. The OH- and Ca++ concentrations in the individual vial were measured at 0, 7, 14 and 30 days. Statistical evaluation was performed with ANOVA and Scheffé tests. The aqueous calcium hydroxide paste showed higher ion concentration at 14 and 30 days. Calcium hydroxide plus propylene glycol showed no significant pH increase. The best solution was EDTA 17 porcentage.ANTIBACTERIAL EFFECTAgar diffusion test Zones of inhibition: NaOCL 2,5 porcentage =21 mm. EDTA 17 porcentage =20mm. CHX 0,2 porcentage =14rnm.Dilution test. Apical and middle root pieces were sterilized by autoclavation and were contaminated with Enterococcus faecalis. The specimens were submerged in vials containing the irrigants and were incubated at 37 degree C. The colony forming units (CFU) were counted at 0, 4, 8, and 24 hours. ANOVA and TUCKEY tests showed the best antibacterial action was NaOCL 2,5 porcentage then CHX 0,25 and finally EDTA 17 porcentage.
